My research is concerned with the toxicology of 2, 3, 7, 8 tetrachlorodibenzo-p-dioxin (TCDD), a compound of great environmental concern. This compound is the most toxic congener of the polychlorinated dioxin class of compounds. It elicits a wide range of toxic effects, including alterations in metabolic pathways. Many of the toxic effects are mediated through a cellular protein known as the aryl hydrocarbon (Ah) receptor. The purpose of our research is to study the involvement of the Ah-receptor in pyruvate carboxylase gene regulation. Pyruvate carboxylase is an enzyme involved in intermediary metabolism leading to the eventual synthesis of ATP. TCDD down regulates the enzyme pyruvate carboxylase which is key to the synthesis of oxaloacetate. We have recently shown that pyruvate carboxylase mRNA and protein levels decrease in C57BL/6J mice injected with TCDD. This decrease appeared dose dependent and was observed between 16 and 192 hours post injection to TCDD. The mechanism(s) responsible for this down regulation of pyruvate carboxylase is under investigation. An understanding of the mechanism may help to understand TCDD wasting that is commonly associated with exposure to this compound. We are also involved in studiees to produce aniline adulterated toxic oils from low erucic acid rapeseed oil (canola oil). This study is a result of a collaboration with the Center for Disease Control and the Government of Spain. The purpose of the study is to produce the actual compounds that caused the death and illness of several thousand people in Madrid, Spain during 1982. A number of triacylglycerol derivatives of aniline have been isolated and purified. These compounds will be used in animal studies to establish their toxicity.
